Drilling Information:

This refers to those holes that do not require "plating-thru", for example mounting holes. However, this creates an extra step in the fabrication process, increasing cost. If you can "make do" with "plating-thru", that is preferred due to cost:

This tells us how many different drill sizes may be used, which affects how many boards can be stacked for drilling and even how often drills may need to be replaced due to wear, factors in quoting your board:

The drill listing is generated by your CAD software and is a listing of all drilled holes on the board grouped by drill size. If you do not have an Excellon drill listing, one will have to be created, which will increase your cost.
Excellon Drill Listing: 

The tool list is generally created when an Excellon drill list is created with your CAD software. It tells us what drill size occupies a particular tool position during the drilling phase:
Tool List: 

The Mechanical Drawing will provide the physical shape and dimensions of the board along with visual drill info and any fabrication requirements. This should also show the relationship of the circuit image to the board outline, that is you should provide a dimension in both X and Y axis from a drilled hole (this will be the "drill origin" used to generate the drill list) to the board outline.
